The t-virus (short for "Tyrant Virus") was a highly adaptable bioweapons-grade pathogen and the main component for the Umbrella Corporation in their biological weapons development program, created from an attempt to weaponize the Progenitor virus for military use and application. The t-virus became the basis for the majority of Umbrella's research. It inherits the genetic modifying properties of the Progenitor virus and drastically improves on them. The very first strain of the t-virus was created by Dr. James Marcus.

Leon Scott Kennedy was an agent for the U.S. Government and a former police officer assigned to Raccoon City. Upon arriving into Raccoon City on his first day as a police officer, he became entangled in the biohazard caused by the t-virus and the illegal activities of its creator, the Umbrella Corporation. After surviving the incident, he was scouted by a U.S. intelligence agent named Adam Benford and recruited into a secret military agency under the direct control of the U.S.

Jill Valentine was a former member of the Special Forces S.T.A.R.S. (Special Tactics and Rescue Service) of the Raccoon City police. She had years of tactical experience and certification and had received training in a training program of the U.S. Army Delta Force where she earned high grades in bomb disposal, making her an extremely competent and experienced member of any unit.

Raccoon City was an urban community located in the American mid-west. At its peak, it had a population of over one hundred thousand. While existing as a town initially, it was thanks to the many donations by Umbrella that Raccoon prospered into a full fledged city in the late 20th century. Following the T-virus outbreak that afflicted the city in September 1998, the entire community was destroyed by an experimental nuclear weapon by the United States government, as part of Mission Code: XX (Bacillus Terminate).
